Возможное влияние физических упражнений на передачу сигналов и воздействие инсулиноподобных факторов роста



Возможные последствия занятий физическими упражнениями па функционирование сигнальной системы ИФР и ИФР-регулируемую физиологию могут проявляться на разных уровнях, включая изменения локальной и общей концентрации ИФР и IGFBP (а также степени протеолиза последних) либо менее заметные воздействия на экспрессию рецепторов ИФР и внутриклеточные процессы. Взаимосвязь между занятиями физическими упражнениями и ИФР, а также их связывающими белками будет рассмотрена в последующих главах, поэтому здесь мы остановимся только па эффектах физической нагрузки на передачу сигналов в системе ИФР. В настоящий момент не имеется никаких данных о влиянии физических упражнений на экспрессию или активность IGF-IR, опосредующего передачу основной массы сигналов от ИФР.

Альтернативная возможность заключается в том, что изменения в экспрессии или активности IR могут модулировать передачу сигналов ИФР. Это может быть и прямое воздействие, если эти изменения затрагивают изоформу рецептора IR-А, поскольку, как говорилось выше, она является функциональным рецептором ИФР-11. Кроме того, изменения экспрессии или активности любой изоформы IR может оказывать опосредованное влияние на передачу сигнала ИФР-1 или ИФР-И через IGF-IR за счет формирования гибридных рецепторов. В то время как существующие данные подтверждают влияние физических упражнений па общее действие инсулина па организм и метаболизм глюкозы в частности (см. обзор Wojtashewski et al., 2002; Zietrach, 2002), степень воздействия двигательной активности на количество или активность IR, особенно у человека, остается неизвестной. Существует одна публикация, где сообщается о влиянии физических упражнений па аутофосфорилированис IR (Youngren et al., 2001), однако в большинстве исследований рассматриваются эффекты на более отдаленные компоненты сигнальных систем, такие, как IRS-1 и IRS-2 (Chibalin et al., 2000; Nagasaki ct al., 2000) и Р13-киназиый путь (Kim ct al., 1998). Вместе с тем эти более отдаленные последствия могут быть неспсцифичными, поскольку они затрагивают сигнальные факторы; которые использует множество других гормонов и ростовых факторов.

Заключение

Несмотря на десятилетия интенсивных исследований, основные аспекты функционирования сигнальной системы ИФР по-прежнему остаются плохо понятными. Главным среди них является роль ИФР-П в физиологии человека. Кроме того, абсолютно не изучено совместное действие ИФР-1 и ИФР-И на клеточном уровне, несмотря на то что большинство тканей в организме человека обычно подвергается комбинации эндокринного, паракринного и часто аутокрипного действия обоих факторов роста. В контексте данной книги комплексность системы ИФР предоставляет широкий спектр мишеней, через которые могут быть опосредованы эффекты воздействия физических упражнений па физиологические и патофизиологические процессы, в регуляции которых принимает участие ИФР.
